This research was conducted to investigate the transfer of β-glucuronidase (GUS) gene to Gerbera jamesonii cv. ‘Royal Soft Pink’ by co-cultivation of leafy petioles explants with Agrobacterium tumefaciens strain EHA101 (pIG121-Hm). This plasmid harboring GUS, hygromycin resistant (hpt) and kanamycin resistant (npt II) genes. After 2-3 days of co-cultivation of leafy petioles explants with Agrobacterium transferred to MS medium containing 4.0 mg L-1 BA, 1.0 mg L-1 TDZ, 0.1 mg L-1 IAA, 10 mg L-1 hygromycin and10 mg L-1 meropenem trihydrate for direct shoot regeneration. Then putative transgenic shoots were derived from petioles cultured on growth MS medium supplemented with 2.0 mg mg L-1 BA, 0. 2 mg mg L-1 NAA, 10 mg L-1 hygromycin and 10 mg L-1 meropenem trihydrate for shoot multiplication. Transformation of plants was confirmed by histochemical staining assay and PCR. Key words: GUS gene, gerbera, direct regeneration, histochemical staining.
