RNA silencing is a method for post-transcriptional gene silencing (PTGS) used frequently in plants. The method is based on RNA degradation process through a sequence spesific nucleotide interactions induced by double-stranded RNA. For genetic improvement of crop plants, RNAi is more efficient and stable in comparison with other methods such as antisense-mediated gene silencing or co-suppression. We have made RNAi constructs using Gateway technology from coding and promoter region of Fructan Exohydrolase. These construct have been introduced into an Agrobacterium C58C1(pMP90) strain for transformation of chicory root cultivar (root cultivar). Transgenic chicory plants containing double stranded RNA of FEHIIa (exohydrolases) could suppress the inulin degradation gene.
