Abstract

Ferula pseudalliacea is a perennial and monocarpic plant that belongs to Apiaceae family and distributed thorough Central Asia to Northern Europe. The main constituents of secondary metabolites in this genus comprises cumarines, sesquiterpene cumarines and disesquiterpen cumarines. Due to high value compounds, this species has been used as medicinal plant by endemic people. Main bioactive constituents in F. pseudalliacea are produced in phenyl propanoids pathway. Phenyl alanine ammonia lyase (PAL) and Cinammate-4-hyrdoxylase important (C4H) genes encodes main enzymes in this pathway. In the present study, we aimed to identify and clone PAL and C4H genes using 3´-RACE technique and studying their expression level in different parts (root, stem, inflorescence and unripe seed) of F. pseudalliacea using Real-Time PCR. We could identify and clone C4H and 3 PAL isoforms using 3´-RACE. On the other hand, expression analysis revealed different transcript levels in leaf, stem, inflorescence and unripe seed. The lowest expression level of PAL isoforms and C4H genes was observed in root. Based on the previous studies on F. pseudalliacea phytochemicals, it seems that there are more isoforms of PAL and C4H in F. pseudalliacea.
