Abstract

Breast cancer is the most common cancer in women worldwide and is a heterogeneous disease with several environmental factors and genetic changes such as polymorphisms. Arg/Pro polymorphism at codon 72 of the P53 as one of the most common site for genetic alteration in human cancers and ABCC1 promoter polymorphism as a risk factor for progressive cancer has been investigated in different populations. In this study, effect of two polymorphisms in 25 tissue samples from women with breast carcinoma and 30 control samples were analyzed by PCR-RFLP with BstUI and HpaII and PCR-SSCP. The distribution of CC, GC and GG genotypes of codon 72 of p53 were 8.00%, 56.00% and 36.00% respectively in patients with cancer and 13.33% CC, 56.67% CG and 30.00% GG in controls. The relative allele frequencies of ABCC1 promoter polymorphism were 54.00% A and 46.00% T in patients and 36.66% for A and 63.34% for T in normal controls. With c2 test there is statistical difference in the distribution of p53 codon 72 and G1666A of ABCC1 polymorphisms between patients and control groups (p<0.05). Study of these samples with PCR-SSCP did not display any new band template. Our results demonstrated that p53 codon 72 and G-1666A of ABCC1 polymorphisms might have a probable role in breast carcinogenesis in range of high population.
